目的鉴定红细胞(RBCs)中有无内皮型一氧化氮合酶(eNOS),探讨在4℃保存条件下,RBCs中的eNOS含量随保存时间变化的规律。方法采用免疫印迹(WB)和免疫荧光技术检测库存红细胞膜标本中有无eNOS;采用逆转录-PCR(RT-PCR)技术eNOS-mRNA库存红细胞中的检测。结果 RBCs中eNOS检测结果呈阳性。灰度分析结果表明:在RBCs保存35 d内,并不能引起eNOS含量的变化;而RBCs中并没有检测到eNOS-mRNA的存在。结论 RBCs含有eNOS;在库存血液35 d的保存期内,RBCs中的eNOS含量保持稳定。另外,在RBCs中未发现有eNOS-mRNA存在。
Objective To identify endothelial nitric oxide synthase(eNOS) existing in human RBCs and to study its changing profile after storage at 4℃ for 15 days.Methods Western blotting and immunofluorescence assays were applied to identify eNOS in the membrane of banked red blood cells.RT-PCR method was used for identification of eNOS-mRNA in banked RBCs.Results eNOS could be detected in RBCs.The gray analyses suggested that the content of eNOS remained constant in banked RBCs at 4℃within 35 d.eNOS-mRNA couldn't be detected in RBCs.Conclusion eNOS exists in human RBCs and the content is stable in banked RBCs stored for 35 d.However,the eNOS-mRNA was not discovered in human RBCs.
