Two differential primers were designed from the sequence of yak's two α-globin genes's intergenic region which was cloned and sequenced, and from the conserved region of human, mouse, goat, horse and cattle's α-globin gene for confirming the order of yak's two α-globin genes. There was no other gene in the intergenic region of yak's two α-globin genes after blasting. The upstream gene was identified as yak's α1-globin gene, while the downstream gene was identified as yak's α2-globin gene after cloning and sequencing. These indicate that the yak's two α-globin genes are located in the chromosome tightly, α1-globin gene in the front and α2-globin gene at the back,
