创伤及高渗透压预处理对蝴蝶兰叶片诱导类原球茎的影响

以蝴蝶兰试管播种苗的顶端第1叶和第2叶叶片为外植体,以1/2 MS+10%椰子乳+NAA 0.1mg·L^-1+TDZ 1.0mg·L^-1+0.5%琼脂为诱导培养基,研究了创伤和高渗透压两类预处理对其诱导类原球茎(protocorm—like body,PLB)的影响。结果表明,两类预处理均对叶片诱导PLB有显著促进作用。在创伤预处理中,当外植体横切2刀及用刀尖扎15次后,外植体的PLB诱导率分别达到66.1%和68.3%,平均每外植体诱导出13.1和15.2个PLB;在高渗透压预处理中,当用0.4mol·L^-1甘露醇溶液和AAM培养基分别浸泡2h后,外植体的PLB诱导率达到55.2%和51.3%,平均每个外植体诱导出13.7和12.9个PLB,而对照的PLB诱导率只有19.5%,平均每个外植体只诱导出3.1个PLB。经上述两类预处理后外植体的PLB诱导率和平均PLB个数均与对照达极显著差异水平,同时,外植体上诱导出PLB的时间分别比对照提前了10d和5d。但来源于第1叶和第2叶的外植体之间PLB诱导结果均没有明显差异。经创伤和高渗透压预处理后诱导出的PLB均能正常分化成苗。

英文摘要:

The first and second leaf from the top of Phalaenopsis amabilis seedlings were used respectively as explants, and 1/2 MS medium ineluding 10% eoeonut milk (CM) , 0. 1 mg·L^-1 α-naphthyl aeetie aeid ( NAA), 1.0 mg·L^-1 thidiazuron (TDZ) and 0. 5 % agar was used to induee PLB, the effeets of wound and high osmotic pressure on PLB (protoeorm-like body) induetion were studied. The results revealed that these two types of pretreatments eould improve PLB induetion remarkably. After the explants were eut transversely twiee or stabbed 15 times on different sites, their PLB induetion rates went up to 66. 1% and 68.3%, the PLB number per explant went up to 13.1 and 15.2. After the explants were ineubated in 0. 4 mol ·L^-1 mannitol solution or AAM medium for 2 hours as high osmotie pressure pretreatments, the PLB induetion rates went up to 55.2% and 51.3% , the PLB number per explant went up to 13.7 and 12.9. While the PLB induetion rate of the eontrol was 19. 5% and the PLB number per explant was 3.1. Compared with the eontrol, the PLB induetion rate and PLB number per explant were both very signifieantly differenee in all the pretreatments. Moreover, after the explants were pretreated by wound or high osmotie pressure, the time of PLB formation was 10 and 5 days shorter respeetively than that of the eontrol. However, there were not obvious differences between the first and second leaf explants in PLB induction. PLB induced from the explants that had been pretreated by wound or high osmotic pressure could develop into plantlet normally.
