目的:观察黄芪注射液对大鼠压力过载性心肌肥厚的逆转作用,并探讨胶原蛋白Ⅰ、Ⅲ在肥厚心肌中的表达变化及黄芪注射液对其的影响。方法:采用缩窄大鼠腹主动脉建立心肌肥厚模型,术后12周开始给药,连续12周。测定大鼠左心室质量指数(LVWI),观察黄芪注射液对大鼠心肌组织病理变化的影响;检测大鼠左心室组织胶原蛋白Ⅰ、Ⅲ的表达。结果:黄芪注射液呈剂量依赖性抑制模型大鼠左心室LVWI的增加,其中高剂量黄芪注射液与卡托普利作用相似。黄芪注射液和卡托普利均能减轻模型大鼠心肌胞外基质过多沉积,改善心肌细胞受压萎缩或空泡变性的程度;均能抑制心衰大鼠左心室组织中胶原蛋白Ⅰ的表达,而胶原蛋白Ⅲ的表达无显著变化。结论:黄芪注射液对压力过载所致大鼠左心室肥厚有抑制作用,其机制可能与下调心肌组织胶原蛋白1的表达有关。
OBJECTIVE: To observe the reverse effect of Radix Astragali injection(AsI) on pressure overload - induced cardiac hypertrophy in rats and discuss the effect of AsI on Collagen Ⅰ、Ⅲ gene expressions in rats with pressure overload - induced cardiac hypertrophy.METHODS: Cardiac hypertrophy was induced by clipping the abdominal aorta in rats.The model rats were administered with drugs at 12 weeks after operation for 12 consecutive weeks.Then the effect of AsI on the pathological change of cardiac hypertrophy was evaluated by left ventricular mass index(LVWI,left ventricular mass/body mass) and the Collagen Ⅰ、Ⅲ protein expressions in left ventricular tissues were determined as well.RESULTS: The increase of LVWI was dose - dependently inhibited by AsI. The effect of high dose AsI was similar to that of Captopril (P 〈 0.001).Both As and Captopril could lessen the excessive deposition of extracellular matrix, ameliorate the atrophy induced by compression or the degree of vacuolar degeneration and inhibit the expressions of Collagen Ⅰ in the left ventricular tissue of the model rats.There was no difference in the expressions of Collagen Ⅲ among all groups.CONCLUSION: AsI can inhibit the development of left ventricular hypertrophy in rats induced by pressure- overload, which may contribute to its down- regulation of collagen I expressions in left ventricular tissues.
