目的观察胰腺癌培美曲塞敏感株Patu8988和耐药株Patu8988/P生物学行为的差异,并探讨其可能的机制。方法取对数生长期的Patu8988、Patu8988/P细胞,分别采用双层软琼脂实验检测细胞集落生成能力、流式细胞仪检测细胞周期分布、Transwell小室检测细胞侵袭及转移能力,采用RT-RCR法、流式细胞仪检测胰腺癌细胞中的CD44mRNA及其蛋白。结果与Patu8988细胞比较,Patu8988/P细胞的克隆形成能力增强,G1期的细胞比例增加,S期减少,而侵袭转移能力减弱,CD44mRNA及其蛋白均表达增高,P均〈0.05。结论 Patu8988/P、Patu8988细胞的生物学行为有不同程度的差异,主要表现在前者集落生成能力增强,侵袭、转移能力减弱等;CD44表达升高可能是造成这种差异的原因之一。
Objective To investigate the biological behavior of pemetrexed-sensitive and pemetrexed-resistant human pancreatic carcinoma cell,and explore its mechanism.Methods The colony formation and its formation rates of pemetrexed-sensitive human pancreatic carcinoma Patu8988 cell and pemetrexed-resistant human pancreatic carcinoma Patu898/P cell were determined by double-layer soft agar colony formation assay,the cell cycle was measured by flow cytometry,the invasion and migration ability of cells was tested by Transwell chamber assay,RT-PCR and flow cytometry method was used to compare the differences in the expression of CD44 mRNA and protein between the two cell strains.Results Pemetrexed-resistant Patu8988 strain had higher ability in colony formation rate but lower ability in invasion and migration ability(P0.05).The expression of CD44 mRNA and protein were higher in pemetrexed-resistant Patu8988 cell.Conclusions There are difference in the abilty of invasion,migration and colony formation rate between pemetrexed-resistant Patu8988/P cell and pemetrexed-sensitive Patu8988 cell.The increase of CD44 expression may be an important raason.
