目的研究广西巴马地区居民血清中叉头框蛋白O3A(forkhead box protein O3A,FOXO3A)基因mRNA及蛋白表达水平的差异,探讨其与家庭聚集性长寿现象的关系。方法于2011年5月,在广西巴马地区抽取具有3-4代直系亲属的家庭,分为长寿家族史组(137例)和无长寿家族史组(91例),测定血清中FOXO3A mRNA及蛋白的表达水平。结果长寿家族史组人群血清FOXO3A mRNA的表达水平是无长寿家族史组的1.21倍,差异有统计学意义(P〈0.05)。长寿家族史组和无长寿家族史组人群血清FOXO3A蛋白的表达水平间比较,差异无统计学意义(P〉0.05)。长寿家族史组居民血清中FOXO3A mRNA的表达水平与其蛋白的表达水平之间未见相关关系(P〉0.05);无长寿家族史组居民血清中FOXO3A mRNA的表达水平与其蛋白表达水平呈负相关(P〈0.05)。结论广西巴马地区居民血清FOXO3A mRNA的表达水平可能与当地家庭聚集性长寿现象有一定关联,尚未发现FOXO3A蛋白水平与家庭聚集性长寿现象的关联性,需进一步扩大样本量进行验证。
Objective To investigate the mRNA and protein level of the FOXO3A gene in residents of Bama,to discuss the correlation between gene expression level and the significant familial aggregation phenomenon of longevity. Methods All the cases in this study were selected from the families which had 3-4 generations of immediate family members and then they were classified as longevity group(137 cases) and non-longevity group(91 cases) according to whether there was a family history of longevity in May 2011. The mRNA and protein level of the FOXO3A gene were detected for 137 residents from longevity group and 91 residents from non-longevity group. Results The mRNA level of FOXO3A in longevity group was 1.21 times as higher as that of non-longevity group(P〈0.05). There was no statistically significant difference on serum FOXO3A protein level between longevity group and non-longevity group(P〈0.05). No correlation between the serum FOXO3A mRNA and its protein level in longevity group was found(P〈0.05); In non-longevity group,negative correlation between the serum FOXO3A mRNA and its protein level was found(P〈0.05). Conclusion The gene expression of FOXO3A in serum may be involved in familial aggregation of longevity phenomenon in Bama country,but the correlation between the protein level of FOXO3A and familial aggregation phenomenon of longevity has not been found in this study and needs further verification.
