目的研究柴胡、葛根药对有效部位分用、合用对脂多糖诱导雄性家兔发热模型的解热作用。方法采用水提-壳聚糖澄清-大孔树脂纯化法,制备了葛根总黄酮有效部位;采用醇提-大孔吸附树脂纯化法和水蒸气蒸馏法从柴胡中提取柴胡总皂苷和柴胡芳香水;将36只雄性家兔随机分为模型组、葛根总黄酮组、柴胡有效部位组、柴葛合用组、对乙酰氨基酚(阳性对照)组及柴胡注射液组,采用脂多糖0.5μg/kg耳缘静脉注射建立家兔发热模型,研究葛根总黄酮及柴胡有效部位配伍前后的退热作用。以平均发热曲线及体温反应指数为指标,观察葛根总黄酮及柴胡有效部位配伍前后的退热作用。结果柴葛合用时,退热效果显著(与模型组比较,P〈0.01),持续时间长(与柴胡组比较,P〈0.05)。结论柴胡葛根有效部位合用时解热效果佳。
AIM To explore the antipyretic effects of single or combining Bupleurum-Pueraria effective fraction on lipopolysaccharide-induced febrile response in male rabbits.METHODS The Pueraria flavonoid fraction was prepared by the method of water extraction-chitosan clarification-macroporous resin purification,and Bupleurum saponins and aromatic water were extracted from Bupleuri Radix by alcohol-macroporous resin purification and steam distillation.Thirty-six male rabbits were randomly assigned to six groups: model group,Pueraria flavonoid group,Bupleurum effective fraction group,Bupleurum-Pueraria compatible group,acetaminophen positive control group,Bupleurum injection group.Fever model was induced by intravenously injecting into lipopolysaccharide(0.5 μg/kg) via ear vein.Referring to average temperature curve and temperature response index,the observation on the antipyretic function before and after the experiment was carried out.RESULTS The result showed that Bupleurum-Pueraria combination had a significantly apparent antipyretic function(compared with the model group,P〈0.01),which also lasted longer(compared with the Bupleurum group,P〈0.05).CONCLUSION The effect of the combination of Bupleurum and Pueraria is much better than that of the single fraction.
