为研究Aβ蛋白显像剂11^C-PIBPET成像技术用于评价阿尔茨海默病大鼠模型中的作用,以9只Aβ1~40注射模型大鼠和10只生理盐水对照大鼠采用HE染色及Aβ蛋白免疫荧光染色来检测大鼠脑内病理改变。11^C-PIB及18^F-FDGPET显像在体观察两组大鼠脑内Aβ蛋白分布及葡萄糖代谢情况。结果发现:模型组海马区出现神经元减少及淀粉样斑块的形成。同时模型组PET显像可见注射侧葡萄糖代谢减低及11^C-PIB摄取增加。对照组仅见轻微神经元损伤及PET所显示的葡萄糖代谢轻度减低。11^C-PIB及18^F-FDGPET显像结合行为学及病理学观察可以作为评价AD模型检验方法,该方法为基础研究及临床诊断AD提供了一种分子成像工具。
This paper investigates the role of 11^C-PIB PET imaging in the evaluation of Alzheimer's disease rat model. 8 AD rat models and 10 control rats were used in 18^F-FDG and 11^C-PIB PET studies. The sections of the brain tissues of both control rats and AD models were stained with HE method and Aβimmunofluorescence. PET imaging shows decreased glucose metabolism and high uptake of 11^C-PIB in the injected side of AD rats. Diffuse neuronal loss and Aβdeposition were detected in the hippocampus and cortex in the AD rats, while those were hardly observed in control rats. It may be concluded that 11^C-PIB and 18^F-FDG PET imaging, combined with the behavioral observation, may serve as standards in evaluating the AD rat models, and as a molecular imaging tool for the basic research and clinic diagnosis of AD.
