为目的:采用正交试验法,优选了茭白鞘叶木质素提取的最佳工艺条件。方法:采用乙酰溴分光光度法测定木质素含量作为考察指标,对影响木质素提取的因素进行了研究。首先,通过单因素试验研究了萃取溶剂、料液比、温度、微波功率、处理时间对脱取效果的影响,然后,利用正交试验法优化最佳提取条件。结果:正交试验结果表明最佳工艺参数为以乙醇硝酸液为萃取液,料液比1:10、微波功率600w、温度90℃下处理20min,在此试验条件下茭白鞘叶木质素一次提取率为66%。
Objective:The optimal conditions for extraction of lignin from sheathing Leaf of Zizania latifolia were investigated by array design. Methods: The content of lignin was determined by the method of Acetyl bromide Spectrophotometry. The effects of extraction solvent, material/liquid ratio, extraction temperature, microwave power, extraction time on the content of lignin in extracts were investigated by one-factor-at-a-time, and the extraction conditions were optimized by orthogonal array design. Results: The optimal extraction conditions were obtained as follows: using ethanol nitric acid solution as extraction solvent, material/liquid ratio 1:10,microwave power 600 W, extraction temperature 90℃, and extraction time is 20 mins, under which the z-latifolia sheathing leaves lignin extraction rate was 66%.
