目的利用低浓度过氧化氢(H2O2)建立兔原代左心房肌细胞氧化损伤模型,研究夹竹桃麻素对心房肌细胞氧化损伤的影响。方法将18只新西兰白兔随机分为对照组、H2O2组和夹竹桃麻素组,分别进行左心房肌原代培养。H2O2组直接在培养好的原代心房肌细胞中加入终浓度为100μmol/L的H2O2培养2h;夹竹桃麻素组以100μg/ml浓度的夹竹桃麻素处理细胞1h,之后加入终浓度为100μmol/L的H2O2共同培养2h。检测氧化和抗氧化指标的变化。结果与对照组比较,H2O2组和夹竹桃麻素组的SOD活力及GSH含量显著减少(P〈0.05),MDA含量和细胞内钙浓度,烟酰胺腺嘌呤二核苷酸磷酸(NADPH)氧化酶亚基p22phox表达均显著升高增加(P〈0.05),H2O2组的线粒体mRNA表达增加。与H2O2组比较,夹竹桃麻素组SOD活力及GSH含量显著增加(P〈0.05),MDA含量和细胞内钙浓度、NADPH氧化酶亚基p22phox和线粒体mRNA表达显著降低(P〈0.05)。结论夹竹桃麻素能够通过抑制NADPH氧化酶,对心房肌细胞的氧化损伤起保护作用。
Objective To study the effect of apocynin on oxidative stress injury of primary atrial cells in rabbits by establishing a primary atrial cell injury model of rabbits with low-consistence H2O2.Methods Eighteen rabbits were randomly divided into normal control group,H2O2 group and apocynin group.All atrial cells were primarily cultured.In H2O2 group,100μmol/L H2O2 was added into cultured primary cells and cultured for 2h.In H2O2 group,100μg/ml apocynin was added into cultured primary cells and cultured for 1h,into which 100μmol/L H2O2 was added and cultured for 2h.Finally,changes in oxidant and anti-oxidant indexes were detected.Results The activity of superoxide dismutase(SOD) and glutathione hormone(GSH) level were significantly lower while the malondialdehyd(MDA) and intracellular free calcium levels as well as the expression level of nicotinamide adenine dinucleotide phosphate(NADPH) p22phox were significantly higher in H2O2 and apocynin groups than in normal control group(P〈0.05).The activity of SOD and GSH level were significantly higher while the MDA and intracellular free calcium levels as well as the expression levels of NADPH p22phox and mitochondrial mRNA were significantly lower in apocynin group than in H2O2 group(P〈0.05).Conclusion Apocynin can protect primary atrial cells against injury by inhibiting NADPH oxidase.
