目的检测趋化因子受体CXCR4/CXCL12信号转导通路在结直肠癌组织、癌旁黏膜组织以及转移淋巴结中的表达情况,分析CXCR4/CXCL12在结直肠癌淋巴结转移中的作用。方法应用Westernblot检测结直肠癌组织中CXCR4/CXCL12表达,免疫组织化学法检测CXCR4/CXCL12在细胞水平的分布。结果结直肠癌组织中CXCRl4/CXCLl2表达水平明显高于正常肠黏膜,分别为正常黏膜的2.8、2.6倍(P〈0.05)。CXCR4/CXCL12表达水平与淋巴结转移有关(P〈0,05),淋巴结组织中CXCR4/CXCL12表达增高,分别为原发肿瘤的2.1倍、1.7倍。CXCR4/CXCL12在结直肠癌组织中表达情况呈线性相关,相关系数为0.523,P〈0.05。结论趋化因子受体CXCR4/CXCL12在原发结直肠癌与淋巴结转移组织中呈高表达,CXCR4信号转导通路可能在结直肠癌淋巴结转移过程中起重要作用。
Objective To investigate the potential role of CXCR4/CXCL12 in carcinogenesis of colorectal carcinoma. Methods In tissue samples from 60 patients with colorectal carcinoma CXCR4 and CXCL12 expression was detected by using Western blot and immunohistochemistry. Results CXCR4/CXCL12 protein levels in colorectal cancerous tissues were 2.8 and 2.6-fold of those in normal mucosa respectively (P 〈 0.05 ). Overexpression of CXCR4/CXCL12 was related to lymph node metastasis in colorectal cancer. The expression levels of CXCR4 and CXCL12 were in lymph nodes were 2.1 and 1.7-fold of those in primary tumor respectively ( P 〈 0.05 ). The expression of CXCR4 had a positive linear correlation with CXCL12 in tumor ( r = 0.523 ,P 〈 0.05 ). Conclusion Abnormal activation of CXCR4/CXCL12 signaling plays an important role in tumorigenesis of colorectal carcinoma.
