目的检测钙调神经磷酸酶(CaN)信号转导通路在结直肠癌组织中的表达,分析CaN成员与结直肠癌临床病理特征的关系。方法应用Westem blot检测40例结直肠癌组织及其邻近正常黏膜中CaN Aα、CaN Aβ、CaNB与bcl-2蛋白表达,免疫组织化学法检测CaN在细胞水平的分布。结果结直肠癌组织中CaN信号传导通路成员CaN Aα、CaN Aβ、CaN B与bcl-2在结直肠癌组织中表达明显升高,分别为正常黏膜的3,45倍、3.67倍、4.31倍与3,18倍(P〈0.05);CaN Aα与CaN郐表达水平与TNM分期较晚(Ⅲ/Ⅳ)有关(P〈0.05),CaN Aα与bcl-2在结直肠癌组织中表达情况呈线性相关,相关系数为0.493,P〈0.05。结论CaN信号转导通路持续活化可能在结直肠癌的发生中起重要作用。
Objective To investigate the potential role of Calcineurin in carcinogenesis of colorectal carcinoma. Methods By using Western blot, the expression of CaN Aα, CaN Aβ, CaN B and bcl-2 was detected in 40 cases of colorectal carcinoma tissue and adjacent normal mucosa. The expression of CaN was assayed by immunohistochemistry. Results The expression of CaN Aα, CaN Aβ CaN B and bcl-2 protein in colorectal cancer tissue was 3.45, 3.67, 4.31 and 3.18-fold that in normal mucosa respectively (P〈0.05). It was found that the overexpression of CaN Aαand CaN Aβ was related to TNM stage in colorectal cancer ( P〈0.05 ). The expression of CaN A had a positive linear correlation with bcl-2 in colorectal cancer tissue ( r=0.493, P〈0.05). Conclusion The abnormal activation of CaN signaling pathway plays an important role in t of colorectal carcinoma.
