目的:筛选并鉴定与老龄肾阳虚证相关的差异表达基因。方法:从前期基因表达谱芯片初步获得的差异表达基因中筛选出25个基因,选取18例肾阳虚证患者和6例正常对照者,采用半定量RT-PCR检测其血液中这25个基因的表达情况,从中鉴定出与老龄肾阳虚证相关的差异表达基因。结果:有8个基因在18例老龄肾阳虚证患者和6例正常人间的表达水平存在明显差异,这8个基因主要涉及细胞周期和凋亡,肿瘤发生以及免疫。结论:老龄肾阳虚证的发生涉及一系列免疫紊乱的复杂过程,推测肾阳虚可能与肿瘤的发生存在某种程度的关联。
Objective:To filter and verify the relevance of the differentially expressed genes of aged kidney-Yang deficiency syndrome.Methods:From the earlier gene expression profile chip,we filtered 25 genes in the differentially expressed genes,18 elderly patients with kidney-Yang deficiency and 6 healthy people were selected,and the semi-quantitative skill of the RT-PCR was used to detect the 25 genes'expressions in the blood so as to verify the differentially expressed genes which are correlated with the aged kidney-Yang deficiency syndrome.Results:There are 8 genes that have the significant differences in expression between the 18 elderly patients with kidney-Yang deficiency and the 6 healthy people.The 8 genes mainly involve the mitotic cycle and apoptosis,cancer and immunity.Conclusion:The aged kidney-Yang deficiency syndrome is related to dysfunction of the immune system.Hence,we can conjecture some relationship between the kidney-Yang deficiency syndrome and the development of tumor.
