目的探讨BRAF基因在子宫内膜异位症异位内膜及在位内膜中的表达。方法采用实时聚合酶链反应技术及免疫组织化学SP法检测20例子宫内膜异位症患者的异位内膜和在位内膜及20例对照组正常子宫内膜中BRAF的表达。结果在位内膜中BRAF mRNA相对表达量(0.0000678970±0.0001145286)明显高于正常内膜(0.0000095431±0.0000064543)(P〈0.05),而异位内膜(0.0000293957±0.0000380469)与在位内膜之间差异无统计学意义(P〉0.05)。异位内膜和在位内膜中BRAF蛋白的阳性表达率和表达强度均明显高于正常内膜(P〈0.05),而异位内膜和在位内膜之间阳性表达率及表达强度的差异无统计学意义(P〉0.05)。结论在位内膜BRAF基因高表达可能在子宫内膜异位症发生中起重要作用,而与病情进展的相关性有待进一步验证。
Objective To investigate the expression of BRAF in ectopic and eutopic endometrium of women with endometriosis.Methods Quanti- tative real-time polymerase chain reaction (RT-PCR) and immunohistochemistry SP method were adopted to measure the BRAF expression in ecto- pic and eutopic endometrium ( n = 20) of patients with endometriosis, as well as normal endometrium ( n = 20) of women without endometriosis. Re- suits Relative expression of BRAF mRNA in eutopic endometrium (0.000 067 897 0±0.000 114 528 6) was significantiy higher than that in con- trol group (0.000 009 543 1±0.000 006 454 3 ) (P 〈 0.05 ), but no difference was found between the ectopic (0.000 029 395 7±0.000 038 066 9 ) and eutopic group (P 〉 0.05 ). The expression rate and intensity of BRAF in ectopic and eutopic endometrium was significantly higher than the con- trol group (P 〈 0.05 ) ; however, there was no significant difference between ectopic and eutopic endometrium (P 〉 0.05 ). Conclusion The high expression of BRAF in eutopic endometrium may play a crucial role in the occurrence of endometriosis, while the correlation with disease progres- sion needs to be further verified.
