目的通过复方地芬诺酯致便秘小鼠排便实验及小肠推进实验,研究商陆皂苷甲(Es A)的泻下作用。方法向复方地芬诺酯致便秘小鼠腹腔注射不同浓度Es A并灌服墨汁,每2 h收集一次粪便,连续收集10 h,记录每只小鼠首粒黑便出现时间,粪便阴干后称量每一时段内的排便量并计算总排便量;向复方地芬诺酯致便秘小鼠腹腔注射不同浓度Es A并灌服墨汁,给药25 min后脱椎处死,测量并计算小肠推进率。结果与模型对照组比较,Es A高、中剂量组小鼠首粒黑便出现时间缩短,总排便量增加,差异有统计学意义(P<0.05)。小肠推进实验中,Es A高、中、低剂量组小鼠小肠推进率与模型对照组比较差异均无统计学意义(P>0.05)。结论排便实验结果表明:高、中剂量的Es A对复方地芬诺酯致便秘小鼠有一定的泻下作用;小肠推进实验结果表明:Es A对复方地芬诺酯致便秘小鼠没有促进小肠推进的作用。
Objective To study the purgative effect of esculentoside A(EsA)via the defecation and small intestine propulsion experiments in the mice of constipation induced by compound diphenoxylate. Meth-ods EsA of different concentrations was given for intraperitoneal injection and ink gavage was applied. The stool was collected once every 2 h,continuously for 10h. The time of the first black stool was recorded in each mouse. The dry stool was weighted in each time point and the total stool amount was calculated. The intraper-itoneal injection of EsA of different concentrations and ink gavage were applied for 25 min,afterwards,the mice were sacrificed by vertebra broken. The small intestine propulsive rate was measured. Results Com-pared with the model control group,in the EsA high - dose and middle - dose groups,the time of the first stool was shortened and the total stool amount was increased,indicating the significant difference(P < 0. 05). In the experiment of small intestine propulsion,the difference in the small intestine propulsive rate in the EsA high - dose,middle - dose and low - dose groups was not significant statistically as compared with the model control group(P > 0. 05). Conclusion The defecation experiment results show that EsA of the high and middle doses acts a certain of purgative effect in the mice of constipation induced by compound diphenoxy-late. The small intestine propulsion experiment results indicate that EsA does not promoted the small intestine propulsion in the mice of constipation induced by compound diphenoxylate.
