目的探讨中药胃肠舒片促进胃肠动力作用机理。方法将50只Wistar大鼠随机分为正常对照组、模型对照组、胃肠舒小剂量组、胃肠舒大剂量组、西沙必利组。除正常对照组外,其余4组均制备胃肠动力障碍动物模型,分别用等容积的受试药液灌胃,连续7d,将各组动物麻醉后选取胃肠标本。免疫组化法观察神经递质Ach及P物质的表达变化,采用图像分析仪测定其肌间神经丛光密度值。结果中药胃肠舒片能明显增加胃肠组织中Ach及P物质分泌,与模型组比较有显著差异。结论中药胃肠舒促进胃肠动力的作用机制,可能是通过增加胃肠道Ach及P物质分泌,Ach及P物质与相应受体结合,增加胞内Ca^2+浓度,Ca^2+信号系统引发一系列生理功能,从而促进胃肠平滑肌的收缩效应。
Objective: To.investigate the effect of Weiehangshu Tablet on gastrointestinal motility. Methods: 50 Wistar rats were divided into 5 groups randomly: normal group, model group, low dosage Weichangshu Tablet group, high dosage Weichangshu Tablet group and cisapride group. In addition to the normal group, the other 4 groups were prepared animal models of gastrointestinal motility disorder. The groups were given the same volume solution of corresponding medicine by gastric perfusion for 7d. The gastrointestinal samples were taken after rats were anesthetized. The expression change of Ach and substance P were determined by immunohistochemical method. The optical density of myenteric plexus were measured with mitrospcregraph. Results: Weichangshu Tablet could effectively increase Ach and substance P in gastrointestinal tract. Compared with model group, there was significant difference. Conclusion: The mechanism of Weichangshu Tablet promoting gastrointestinal motility may be that can increase Ach and substance P, which act on corresponding receptors and enhance the Ca^2 + concentration then the Ca^2+ signal system cause gastrointestinal smooth muscle contraction.
