目的:探讨2种肝癌细胞BEL-7402细胞及BEL-7402/FU细胞中差异表达的miRNA并对其基因表达谱进行生物学信息分析。方法:采用TRIzol一步法提取BEL-7402细胞及BEL-7402/FU细胞的总RNA,并纯化mRNA,反转录合成荧光分子Hy3标记的cDNA探针,与基因芯片杂交;采用Genepix Pro 6.0图像分析软件对芯片图像进行分析,把图像信号转化为数字信号,然后以差异为2倍的标准来确定差异表达基因。结果:在333个基因表达谱的筛选中,发现有2个基因表达水平显著上调,331个基因表达水平显著下调。结论:miR-122,miR-195等基因组对表达肝癌细胞耐药基因表达谱有显著的影响,可能参与肝癌耐药的发生、发展。
Objective: To screen the differential miRNA expression between BEL-7402 and BEL-7402/FU cell lines.Methods: The total RNA of BEL-7402 and BEL-7402/FU cells was isolated and reversely transcribed to synthesize the cDNA probes labeled with 2 kinds of fluorescences(Hy3).The cDNA was subjected to microarray analysis by GeneChip.Genes differentially expressed between BEL-7402 and BEL-7402/FU cells were identified by Genepix Pro 6.0 software with the standard of 2-fold differential expression.Results: The microarray results revealed that 2 genes were up-regulated and 331 genes were down-regulated among screened 333 genes.Conclusion: The changes in expression of some miRNAs(such as miR-122,miR-195) play a role in regulation of drug resistance gene expression in hepatoma cells,which might be related to drug resistance in the development of liver cancer.
