目的 探讨马鞭草总苷(TGV)对大鼠慢性非细菌性前列腺炎(CNP)的影响。方法 将10月龄健康Wistar大鼠50只前列腺背侧叶注射25%消痔灵造成CNP模型,随机分为模型组、前列康(1000 mg/kg)组及TGV高、中、低剂量(120、60、30 mg/kg)组,每组10只,另以10只正常大鼠作为正常组。自造模后第8天起,各给药组分别灌胃给予相应药物,模型组和正常组灌胃等体积生理盐水,每日1次,连续22 d。给药结束后测定各组大鼠前列腺湿重,计算前列腺指数,检测前列腺液中白细胞数和卵磷脂小体密度,放免法测定前列腺组织匀浆中白介素-2(IL-2)、白介素-1β(IL-1β)、肿瘤坏死因子-α(TNF-α)的含量,HE染色观察前列腺组织的病理学变化。结果 与模型组比较,TGV高、中、低剂量组大鼠前列腺湿重、前列腺指数、前列腺液中白细胞数及前列腺组织IL-2、TNF-α、IL-1β水平降低,前列腺液中卵磷脂小体密度升高,差异有统计学意义(P〈0.01或P〈0.05)。形态学观察显示:TGV对CNP大鼠前列腺组织病理学改变有一定的改善作用。结论 TGV对消痔灵诱导的CNP大鼠具有良好的抑制作用,其作用机制可能与TGV下调前列腺组织IL-2、TNF-α、IL-1β水平有关。
Objective To investigate the effect of total glucosides of Verbena officinalis L. (TGV) on rat model for chronic nonbacterial prostatitis (CNP). Methods CNP model was established by injection of 25% Xiaozhiling through dorsal part of prostate for 50 healthy Wistar rats aged 10 months, the rats were randomly divided into 5 groups, which were model group, Qianliekang (1000 mg/kg) group, high-dose, middle-dose, low-dose (120, 60, 30 mg/kg) of TGV group, with 10 rats in each group. Besides, 10 normal rats were recruited as normal group. Since the 8th day of modeling, the rats of each administered group have been treated with corresponding medicine by intragastric administration, the model group and normal group were given same volume of normal saline by intragastric administration, once daily, for 22 consecutive days. After delivery, the wet weight and prostate index in rat prostate were measured and calculated, and the number of white blood cells and the density of lecithin body in prostatic fluid were observed, the levels of interleukin-2 (IL-2), interleukin-1β (IL-1β) and tumor necrosis factor-α(TNF-α in prostate tissue of rats were detected by radioimmunoassay, and the rat prostate tissue morphology changes were observed by light microscopy after treatment. Results Compared with model group, the wet weight of prostate, the prostate index, the number of white blood ceils in prostatic fluid, and the levels of IL-2, IL-1β, TNF-α in prostate tissue of the high-dose, middle-dose, low-dose of TGV group were decreased, and the density of lecithin body in prostatic fluid was increased, the differences were statistically significant (P 〈 0.01 or P 〈 0.05). Morphological observation indicated that TGV could improve pathological conditions of prostate tissue of CNP rats to some extent. Conclusion TGV has certain therapy effect on CNP rats induced by Xiaozhiling, which may be related to its regulating the levels of IL-2, IL-1β and TNF-α.
