研究3,4-二羟基氰苯对酪氨酸酶的抑制作用和对几种微生物的效应.结果表明:3,4-二羟基氰苯对蘑菇酪氨酸酶活力有明显的抑制作用,其效应除了对稳态酶活力有较强的的抑制作用外,对酶促反应的迟滞时间也有延长作用.10.0μmol/L的3,4-二羟基氰苯可使稳态酶活力下降53.5%,并使迟滞时间从18s延长到225s.3,4-二羟基氰苯对大肠杆菌、金黄色葡萄球菌、枯草芽孢杆菌3种细菌和真菌白色假丝酵母均有明显的抑制作用,其对细菌的最小抑菌浓度(MIC)和最小杀菌浓度(MBC)均分别为500和1000μg/mL,对白色假丝酵母的MIC和MFC分别为1000和2000μg/mL.本研究为3,4-二羟基氰苯在酪氨酸酶抑制剂中的应用提供实验理论依据.
The inhibitory effects of 3,4-dihydroxybenzonitrileon on the activity of mushroom tyrosinase and antibacterial activities against some bacteria and fungi were studied. 3,4-Dihydroxybenzonitrile was found to inhibit the monophenolase activity of mushroom tyrosinase. The lag time of the enzyme for oxidation of L-tyrosine extended and the steady-state rate of monophenolase decreased. When the concentration of 3,4-dihydroxybenzonitrile reached 10.0μmol/L, the steady-state rate of the enzyme dropped by 53.5 % of the total activity,and the lag time extended from 18 sec to 225 sec. The inhibition of 3,4-dihydroxybenzonitrile on some bacteria such as E .coli, B. subtilis, S. aureus and fungi such as C. albicans was studied. The values of MIC and MBC of 3,4-di- hydroxybenzonitrile to E. coli,B, subtilis,St, aureus were determined to be 500 and 1 000μg/mL, respectively,and the values of MIC and MFC to C. albicans were 1 000 and 2 000μg/mL,respectively. This study gave experimental and theoretical support for 3,4-dihydroxybenzonitrile used as inhibitor of tyrosinase and antimicrobial agent.
