为检测不同耐药水平大肠杆菌的acrA和marA基因转录水平,阐明t耐药大肠杆菌株药物敏感性变化与acrA和marA的mRNA水平之间的关系。用定量RT—PCR的方法比较多重耐药菌株SEMR、单药耐药菌株SEICI和SEICH以及质控株ATCC25922的acrA和marA的mRNA水平。结果表明,acrAmRNA水平,SEMR均是SEICI、SEICH和ATcC25922的16倍;marA mRNA水平,SEMR是SEICH的4倍、SEICl和ATCC25922的8倍;SEICI和SEICH的acrA mRNA和marA mRNA水平与ATCC25922无明显差异;同一菌株acrA mRNA和marA mRNA水平的变化保持了较为稳定的一致性。因此认为,大肠杆菌多重耐药菌株SEMR的acrA mRNA和marA mRNA水平与其耐药水平存在相关性。
To inspect the transcriptional level of acrA and marA gene of different resistance level in Escberichia coli, clarify the relationship between acrA mRNA, marA mRNA and resistance level in the Escherichia coli resistance strains. The quantify RT-PCR was used to compare the level of acrA mRNA, marA mRNA of muti-drug resistance strain SEMR, single-drug resistance strain SEICI and SEICH with control strain STCC25922. The level of acrA mRNA, the SEMR is SEICI, SEICH and the ATCC25922 of 16 times; the level of marA mRNA, the SEMR is SEICH of 4 times, SEICI and ATCC25922 of 8 times; the level of acrA mRNA and marA mRNA of SEICI and SEICH have no significant difference with ATCC25922; the change of acrA mRNA and marA mRNA in the same strain kept with stable variety.There were relativity in the level of acrA mRNA and marA mRNA of muti-drug resistance strain SEMR in E. coli with the level of its drug resistance.
