目的研究胰腺癌肿瘤干细胞体外增殖能力及其对化疗药吉西他滨的耐受性。方法以CD44^+CD24^+ESA^+为表面标记,用流式细胞仪在人胰腺癌Panc-1细胞系中分选肿瘤干细胞,接种在96孔板中,用CCK8法检测其体外增殖能力,描制生长曲线。用吉西他滨分别干预普通肿瘤细胞和肿瘤干细胞,分析化疗药刺激对两种细胞生长的影响,并计算IC50值。结果CD44^+CD24^+ESA^+阳性细胞生长速度明显慢于Panc-1细胞。吉西他滨抑制48h,Panc-1细胞IC50值为982μg/甜,CD44^+CD24^+ESA^+细胞Bo值为1981倒ml;抑制72h,Panc-1细胞IGo值为670μg/ml,CD44^+CD24^+ESA^+细胞IC50值为1484μg/ml。结论肿瘤干细胞在体外培养中表现缓慢生长和对化疗药物耐受的特性。
Objective To study the proliferation and drug-resistence ability of pancreatic cancer stem cell in vitro. Methods The pancreatic cancer stem cells were isolated from CD44+ CD24+ ESA+- labeled human cell line Panc-1 with flow cytometry, which then were inoculated in 96-well plates. The in vitro proliferation ability of cancer stem cells was tested and growth curve was drawn by CCK8 staining. Gemcitabine was used to intervene common tumor cells and tumor sten cells, respectively. The effect of gemcitabine on cell growth was investigated and the IC50 was calculated. Results The growth of CIM4^+ CD24^+ ESA^+ positive cells was lower than that of Pane-1 cells. The ICs0 at 48 h of coculture was 982 μg/ml for Panc-1 cells and 1981μg/ml for CIN4^+ CD24^+ ESA^+ positive cells, which at 72 h was 670 μg/ml for Panc-1 cells and 1484μg/ml for CD44^+ CD24^+ ESA^+ positive cells. Conclusion Cancer stem cells in vitro manifest the characteristics of declined proliferation and highly drug-resistence.
