目的:研究麦冬皂苷D对HUVEC凋亡相关分子的影响,探讨其作用机制。方法:用H20:构建凋亡模型,运用MTT及流式细胞仪检测细胞活性,激光共聚焦方法测定粒体膜电位和钙离子浓度。结果:麦冬皂苷D可以稳定线粒体膜电位。减少钙离子内流,增加细胞的活力。结论:麦冬皂苷D对HUVEC有一定的保护作用。
Objectives: To investigate the effect of ophiopogonin D on the apoptosis of vascular cell inducted by H2O2, and to reveal the molecular mechanism, Method: Apoptosis model of HUVEC was established by inducing with H2O2. The activity of HUVEC cell was detected by MTT assay with flow cytometry. Mitochondrial membrane potential and Ca^2+ concentration in HUVEC was determined by laser scanning confocal microscope. Results: The presence of ophiopogonin D increased the activity of HUVEC cell by stabilizing mitochondrial membrane potential and decreasing intracellular Ca^2+. Conclusions: Ophiopogonin D protects the vascular endothelial cell against apoptosis induced by H2O2.
