目的:研究麦冬正丁醇提取部位对H2O2造模的人脐静脉血管内皮细胞(HUVEC)凋亡相关基因和信号转导通路调控,探讨其对HUVEC保护作用的分子机制.方法:用H2O2构建HUVEC凋亡模型,运用MTT检测细胞增殖情况,流式细胞仪检测凋亡相关基因Bcl-2表达,免疫组化方法观察NF-κB表达情况.结果:麦冬正丁醇提取部位可以减少H2O2造模的HUVEC的凋亡,促进Bcl-2的表达,并能降低NF-κB的表达.结论:麦冬正丁醇提取部位可以通过NF-κB路径,促进凋亡抑制基因Bcl-2的表达,从而起到抗HUVEC凋亡的作用.
Objective: The role of n-butanol extract against the apoptosis of vascular endothelial ceils induced by H2O2 and the expression of apoptosis-related gene and signal transducdon factors were investigated to reveal the molecular mechanism on HUVEC protection. Methods: Apoptosis model of HUVEC was established by inducing of H2O2. MTT assay was used to measure the proliferation of the ceils. Immunohistochemisry staining and flow cytometry were used to detect the expression of the correlated apoptosis genes bcl-2 and NF-κB. Results: n-Butanol extract decreased the apoptosis of VEC induced by H2O2 with the up-regulation of Bcl-2 expression and down-regnladon of NF-κB expression. Conclusions: n-Butanol extract protect the vascular endothelial cells against apoptosis induced by H2O2 by regulating the expressions of bd-2 and NF-κB genes.
