目的:观察重组人表皮细胞生长因子等多种细胞因子对滋养层细胞表达基质金属蛋白酶-2,9的影响。方法:将分离纯化的原代培养早孕滋养层细胞分别给予10 ng/mL重组人表皮细胞生长因子、白细胞介素-1α,10及5ng/mL肿瘤坏死因子-α处理48 h后,采用RT-PCR方法检测基质金属蛋白酶-2和9的转录表达。结果:重组人表皮细胞生长因子、肿瘤坏死因子-α增强滋养层细胞中基质金属蛋白酶-2的表达,而白细胞介素-1α,10则对早孕滋养层细胞中基质金属蛋白酶-2的表达具有抑制作用;重组人表皮细胞生长因子亦增强滋养层细胞中基质金属蛋白酶-9的表达,白细胞介素-10对早孕滋养层细胞中基质金属蛋白酶-9的表达同样具有抑制作用,而肿瘤坏死因子-α则对早孕滋养层细胞中基质金属蛋白酶-9的表达无影响。结论:重组人表皮细胞生长因子可通过诱导基质金属蛋白酶-2,9的表达对滋养层细胞侵入起促进作用;白细胞介素-10则通过抑制基质金属蛋白酶-2,9的表达对滋养层细胞的侵袭性起负调节作用。肿瘤坏死因子-α和白细胞介素-1α亦可能通过对不同类型基质金属蛋白酶表达的影响参与滋养层细胞的侵入调节。
Objective To investigate the effect of different cytokines on the expression of matrix metalloproteinase-2 and-9 in isolated trophoblasts.Methods After 48-hour incubation with 10 ng/mL epidermal growth factor,interleukin-1α,interleukin-10 and 5 ng/mL tumor necrosis factor-α respectively,the expressions of matrix metalloproteinase-2 and-9 in trophoblasts isolated from the first trimester villi were detected with the method of reverse transcription polymerase chain reaction.Results Epidermal growth factor and tumor necrosis factor-α induced while interleukin-1α and-10 reduced matrix metalloproteinase-2 expression in trophoblasts.Epidermal growth factor also increased the expression of matrix metalloproteinase-9.However,interleukin-10 suppressed matrix metalloproteinase-9 expression.Tumor necrosis factor-α exerted no effect on matrix metalloproteinase-9 expression in trophoblasts.Conclusion Epidermal growth factor can promote the invasion of trophoblast by inducing matrix metalloproteinase-2 and-9 expressions.Interleukin-10 can reduce trophoblast invasion by down-regulating matrix metalloproteinase-2 and-9 expressions.Tumor necrosis factor-α and interleukin-1α might also be involved in the regulation of trophoblast invasion by influencing the activities of different proteinases.
