目的:观察表皮生长因子等多种细胞因子对滋养层细胞表达基质金属蛋白酶等侵袭相关基因的影响。方法:将分离纯化的原代培养早孕滋养层细胞分别予10ng/mL的表皮生长因子、白细胞介素1α、白细胞介素10,5ng/mL的肿瘤坏死因子α处理48h后,采用RT-PCR方法检测肿瘤转移相关蛋白1和肿瘤转移抑制基因的转录表达,用ELISA检测基质金属蛋白酶-2和黏附分子CD44v6蛋白的表达。结果:表皮生长因子使早孕滋养层细胞中肿瘤转移相关蛋白1mRNA的表达减少,肿瘤转移抑制基因KAI1mRNA表达增加,并诱导基质金属蛋白酶-2和黏附分子CD44v6蛋白的表达;肿瘤坏死因子α可使早孕滋养层细胞中肿瘤转移相关蛋白1mRNA表达增加,肿瘤转移抑制基因KAI1mRNA表达降低,而对早孕滋养层细胞中基质金属蛋白酶-2和黏附分子CD44v6蛋白的表达影响不明显;而白细胞介素1α、白细胞介素10抑制早孕滋养层细胞中基质金属蛋白酶-2和黏附分子CD44v6蛋白的表达。结论:表皮生长因子、白细胞介素1α、白细胞介素10和肿瘤坏死因子α可能通过改变基质金属蛋白酶-2、肿瘤转移相关蛋白1、肿瘤转移抑制基因KAI1、黏附分子CD44v6等侵袭相关基因的表达,在滋养层细胞侵入的调节中扮演不同的角色。
Objective To investigate the effect of different cytokines on the expression of invasion-associated genes in the first trimester trophoblasts.Methods After 48-hour incubation with 10 ng/mL EGF,IL-1α,IL-10 and 5 ng/mL TNF-α respectively,the expressions of MTA1 and KAI1 mRNA in trophoblasts isolated from the first trimester villi were detected with RT-PCR,and the expressions of matrix metalloproteinase-2 and adhesion molecule CD44v6 protein were detected with cell ELISA method.Results EGF induced KAI1 mRNA,MMP-2 and CD44v6 protein expressions and reduced MTA1 mRNA expression in the first trimester trophoblasts.TNF-α induced MTA1 and reduced KAI1 mRNA expression,and showed no effect on matrix metalloproteinase-2 and CD44v6 protein expression in trophoblasts.IL-1α and IL-10 suppressed matrix metalloproteinase-2 and CD44v6 protein expression in trophoblasts.Conclusion EGF,IL-1α,IL-10 and TNF-α might play different roles in regulating the first trimester trophoblast invasion through changing the expression of invasion associated genes such as MTA1,KAI1,matrix metalloproteinase-2 and CD44v6.
