目的:分离培养正常早孕期人滋养层细胞并观察白细胞介素-10,15对其形态的影响。方法:用改良组织块培养法分离正常早孕期人滋养层细胞,用相差显微镜、Giemsa染色、免疫细胞化学法分析其形态。另用10ng/mI。白细胞介素-10,15分别处理细胞48h后,观察其对滋养层细胞形态变化的影响。结果:分离获得纯度〉95%的滋养层细胞,特异表达细胞角蛋白7,并可能有限传代培养。经白细胞介素-15处理的细胞形态变圆,突起或伪足消失,胞浆变厚、深染;而白细胞介素-10处理使细胞胞浆变薄、淡染,周围突起或伪足拉长。结论:改良组织块培养法可有效获得早孕期人细胞滋养层细胞,白细胞介素-10,15可改变滋养层细胞形态。
Objective To isolate human first trimester cytotrophoblasts and observe the effect of IL-10 and IL-15 on the morphology of primarily cultured cells. Methods Human first trimester cytotrophoblasts were isolated with modified tissue block culture method. Before and after 48 hours incubation with 10 ng/mL IL-10 or IL-15, the cells morphology was observed by discrepancy microscopy, Giemsa dye and immunocytoehemistry. Results Cytotrophoblasts were harvested with purity rate 〉95% and cytokeratin 7 positive. The cells could be transferred with limited passages. IL-10 and IL-15 could change the morphology of trophoblasts differently. Conclusion By modified tissue block culture method, human trophoblasts of the first trimester pregnancy can be harvested, and the morphology of trophoblasts can be affected by IL-10 and IL-15.
