目的探讨人eag相关基因(HERG)通道阻断剂cisapride对胃癌细胞凋亡的影响及机制。方法应用HERG通道阻断剂cisapride干预胃癌细胞的HERG电流后,采用流式细胞仪检测胃癌细胞周期分布及细胞凋亡情况;采用透射电镜技术观察胃癌细胞超微结构的变化。结果 cisapride可以阻断胃癌细胞中的HERG电流,阻止胃癌细胞由G1期进入S期;可以诱导胃癌细胞出现凋亡,并呈现典型的凋亡细胞的形态学特征。结论 HERG通道阻断剂cisapride抑制胃癌细胞G1/S期转化,促进胃癌细胞凋亡。
Objective To explore the effects of human eag related gene ( HERG) channel blocker cisapride on the apoptosis of gastric cancer cells and the mechanism .Methods After interfereing HERG current by HERG channel bloc-ker, the cell cycle distribution and opoptosis of gastric cancer cells were investigated by flow cytometry .The ultrastructure of gastric cancer cells was observed under transmission electron microscope .Results Cisapride blocked the HERG current in the gastric cancer cells;HERG channel blocker inhibited gastric cancer cells entering into S phage from G 1 phage and in-duced the apoptosis of gastric cancer cells .Typically morphological changes of apoptosis cells were observed under trans-mission electron microscope .Conclusion HERG channel blocker cisapride can inhibit progression of G 1/S and promote the apoptosis of gastric cancer cells .
