目的:观察CD99基因转染对霍奇金淋巴瘤L428细胞株表型、细胞周期与凋亡的影响。方法:L428细胞分为3组:转染CD99基因组、转染空质粒组及空白对照组。采用免疫细胞化学方法检测3组细胞CD15、CD30与CD99的表达,并用流式细胞术分别检测细胞周期和细胞凋亡。结果:与其他2组相比,转染CD99基因组L428细胞CD15、CD30表达消失,但可检测到CD99的表达。3组细胞周期分布存在差异,转染CD99基因组细胞阻滞于G2/M期(F=24.85,P〈0.001)。3组细胞凋亡率相比,差异有统计学意义(F=45.30,P〈0.001),其中转染CD99基因组细胞凋亡率为(12.7±4.6)%,高于转染空质粒组的(4.0±0.9)%和空白对照组的(3.6±1.6)%(P〈0.05)。结论:CD99基因可能在霍奇金淋巴瘤的发生中起重要作用。
Aim:To observe the effects of CD99 gene transfection on the phenotype,cell cycle distribution and apoptosis of L428 cells.Methods:L428 cells were allocated into 3 groups:cells transfected with CD99 gene,those with empty plasmid,and L428 cells without treatment.Immunocytochemistry was applied to detect CD15,CD30,and CD99 expression.Cell cycle analysis and apoptosis were determined by flow cytometry.Results:Compared with the other 2 groups,L428 cells transfected with CD99 showed no expression of CD15 or CD30,but that of CD99.In addition,cell cycle arrested at G2/M(F=24.85,P0.001)and increase of cellular apoptosis was detected in L428 cells transfected with CD99(F=45.30,P0.001).Conclusion:CD99 may play an important role in the occurrence of Hodgkin lymphoma.
