以内生解淀粉芽孢杆菌为试材,采用单因素和正交实验,通过比较发酵液的生物量及其对棉花黄萎病菌的抑菌活性,对内生解淀粉芽孢杆菌(Bacillus amylolique faciens)X-278液体发酵条件进行了优化。结果表明:内生解淀粉芽孢杆菌X-278培养基各组分的最佳配比为葡萄糖1.5%、花生饼粉29/6、硫酸铵0.5%、碳酸钙0.1%、硫酸镁0.5%、氯化钠0.5%;最佳发酵培养条件为初始pH7,培养温度34℃,转速180r/min,种龄为24h,装液量150ml/500mI。三角瓶,接种量2%,发酵时间为72h。优化后的发酵液中活菌数量达到1.5×10^10CFU/mL,对棉花黄萎病菌的抑菌圈直径为33.4mm。
Taking Bacillus amyloliquefaciens as test material, using single factor and orthogonal experiments,and through comparing biomass of fermentation broth and bacteriostatic activity against Verticillium dahliae Kleb,the fermentation conditions of endogenous Bacillus amyloliquefaciens X- 278 optimization of the medium and conditions: 1.5% glucose, 2% were optimized.
