以采自海南省福山香蕉园香蕉枯萎病发生地和正常种植园的土壤样品为试验材料,对这两类土壤的细菌多样性进行研究。通过构建2种土样的细菌16S rDNA基因文库,利用限制性内切酶HhaI对随机克隆进行酶切筛选,测定代表克隆的16S rDNA序列,并对2种土样的细菌种群进行了系统发育分析比较。结果表明正常香蕉种植区的土壤样品的细菌多样性较为丰富,其中变形菌门、厚壁菌门、酸杆菌门为主要细菌类群;而香蕉枯萎病发生地土壤以放线菌门和厚壁菌门为主要细菌类群。
Soil samples were collected from banana wilt occurred and normal-growing areas from Fushan,Hainan to study the microbial community diversity.After constructing the 16S rRNA gene library of the two kinds of soils,the library was screened with HhaI and sequenced.It revealed that there exited difference of microbial diversity in the the soils of banana and banana wilt.Phylogenetic analysis on bacterial community showed that members of Proteobacteria,Acidobacteria,Firmicutes,were the dominant in the soil of nomal banana,while members of Firmicutes and Actinobacteria were the dominant in the soil of banana wilt.The microbial community diversity of the nomal soil of banana was richer than that of wilt banana.
