以九州虫草为试材,采用三波长-紫外分光光度法,测定了九州虫草超临界流体萃取物中的总黄酮含量。结果表明:以芦丁为标准品,选定244、272、302nm为测定波长,标准曲线回归方程为y=0.025 3x-0.090 4(r2=0.999 7);在浓度16-56μg·mL-(-1)内,△A与浓度之间呈良好的线性关系,表明该方法稳定性好,回收率为94.80%-97.75%,精密度RSD〈0.67%(n=6),超临界提取获得的虫草黄酮含量为1.89mg·g-(-1)。
Cordyceps kyushuensis was used as test material,three-wavelength ultraviolet spectrophotometry was used for determining the content of total flavones in Cordycepskyushuensiswhich was extracted by Supercritical fluid method(SFE).The results showed that regression equation ofΔA rutin standard concentration was obtained:y=0.025 3x-0.090 4 with a relation coefficient r2=0.999 7.Rutin concentrations in 16-56μg·mL-(-1) withΔA obeyed the linear relation measured at wavelengthλ1=302nm,λ2=272nm andλ3=244nm.It indicated that the method had a good stability.The recovery was 94.80%-97.75%;the relative standard deviation was less than 0.67%(n=6).The content of total flavones extracted fromCordyceps kyushuensis by SFE was 1.89mg·g-(-1).
