目的构建肿瘤凋亡基因Fas小干扰RNA(siRNA)表达质粒,研究其对人肺腺癌A549细胞Fas基因表达的阻抑作用,探索Fas/FasL途径在肺癌转移中的作用机制。方法设计有小发夹结构的3条Fas siRNA对应模板DNA序列,退火处理后克隆至pGCsi-U6质粒,构建重组质粒pSi-Fas。将pSi-Fas转染人肺癌A549细胞,采用Western blot检测Fas表达。结果酶切及测序证实质粒pSi-Fas构建成功,可显著抑制A549细胞Fas蛋白表达。结论成功构建的Fas siRNA表达质粒pSi-Fas能抑制人肺癌A549细胞Fas蛋白表达。
Qbjective To clone the recombinant plasmid of small interfering RNA(siRNA) against Fas gene and to evaluate its activity of inhibiting the Fas expression in human lung carcinoma A549 cells, and explore the mechanism of Fas/FasL pathway on lung cancer metastasis. Methods Three Fas siRNA template DNA sequences were designed and synthesized. The annealed siRNA template was inserted into pGCsi-U6 plasmid, which was transfected into A549 cells with limpofectin 2000,The effect of pSi-Fas on the Fas expression of human lung carcinoma A549 cells was detected by Western blot. Results It was confirmed by restrictive enzyme digestion and sequence analysis that the recombinant plasmid was cloned and the aim sequence was obtained. The Fas expression was inhibited at protein levels by the recombinant pSi-Fas. Conclusion Fas siRNA expression plasmid pSi-Fas successfully constructed,which can inhibit the expression of Fas protein in human lung carcinoma A549 cells.
