目的观察侯氏黑散方中的风药、补虚药对凋亡蛋白半胱氨酸天冬氨酸蛋白酶(Caspase-3)及DNA修复蛋白多聚腺苷二磷酸核糖聚合酶(PARP)在缺血脑组织表达的影响。方法线栓法制备大脑中动脉栓塞(MACO)模型,缺血动物于术后随机分成模型组、风药组、补虚药组、风药补虚药联用组。分别在术后6、24、48h3个时间点取材,运用免疫组织化学方法结合图像分析技术,检测大鼠缺血侧海马CAl区、顶叶皮层、纹状体凋亡蛋白Caspase-3及DNA修复蛋白PARP的表达。结果缺血6h,风药可降低海马CAI区、顶叶皮层凋亡蛋白Caspase-3的表达,下调纹状体PARP的表达;补虚药可上调大鼠海马CAI区、顶叶皮层、纹状体PARP的表达。缺血24h,风药可明显降低海马CAI区、顶叶皮层Caspase-3的表达。缺血48h,风药可明显降低顶叶皮层、纹状体凋亡蛋白Caspase-3的表达,下调纹状体PARP的表达,补虚药可明显上调纹状体PARP的表达。结论侯氏黑散方中风药主要通过不同时间、不同部位选择性下调Caspase-3的表达而补虚药则主要通过上调DNA修复蛋白PARP的表达,促进脑细胞的自我修复功能以防止脑细胞凋亡的发生。
Abstract:Objectives To observe the influences of wind-dispelling and tonifying Chinese medicinals in Houshihei San on the expressions of Caspase-3 and poly-ADP-ribose polymerase (PARP) in rat with cerebral iscbemia. Methods The model of middle cerebral artery occlusion (MCAO) was established by using thread approach, and then the rats were randomly divided into model group, wind-dispelling group, tonifying group and combining group. The expressions of Caspase-3 and PARP in hippocampus CA1 zone, parietal cortex and corpus striatum of ischemia side were detected by using immunohistoehemistry technique and imagine analysis at three time points (6 hours, 24 hours and 48 hours after the operation ). Results After ischemia for 6 hours, the wind-dispelling Chinese medicinals reduced the expression of Caspase-3 in CA1 zone and parietal cortex and regulated downwards PARP expression in corpus striatum. The tonifying Chinese medicinals regulated upwards PARP expression in CA1 zone, parietal cortex and corpus striatum. After ischemia for 24 hours, the wind-dispelling Chinese medieinals reduced significantly the expression of Caspase-3 in CA1 zone and parietal cortex. After isehemia for 48 hours, the wind-dispelling Chinese medicinals reduced significantly the expression of Caspase-3 in parietal cortex and corpus striatum and regulated downwards PARP expression in corpus striatum, and the tonifying Chinese medicinals regulated upwards significantly PARP expression in corpus striatum. Conclusion The winddispelling Chinese medicinals in Houshihei San can regulate downwards the expression of Caspase-3 selectively at different time points and positions, and tonifying Chinese medicinals can regulate upwards PARP expression to improve the self-rehabilitation of brain cells to prevent apoptosis.
