目的采用析因实验设计研究天然维药没食子鞣质及没食子鞣质联合氟化钠对变形链球菌浮游及生物膜状态下乳酸脱氢酶活性的影响。方法用含2%蔗糖的脑心浸液液体培养基分别培养浮游和生物膜状态下的变形链球菌,根据析因实验的分组将配置好的没食子鞣质、氟化钠、没食子鞣质联合氟化钠加入相应的菌液中厌氧培养18 h。通过还原性辅酶Ⅰ氧化法测定乳酸脱氢酶活性。使用SPSS 17.0软件包对实验结果进行析因分析,对药物抑制率进行单因素方差分析。结果生物膜状态下乳酸脱氢酶的活性较浮游状态下高;没食子鞣质、氟化钠、没食子鞣质联合氟化钠对浮游和生物膜状态下的乳酸脱氢酶活性均有抑制作用,浮游状态下乳酸脱氢酶对药物作用更敏感;没食子鞣质对乳酸脱氢酶活性的抑制作用最为显著,抑制率没食子鞣质〉氟化钠〉没食子鞣质联合氟化钠。结论没食子鞣质可能是通过抑制乳酸脱氢酶活性抑制变形链球菌产酸,从而达到防龋的作用。
Objective Based on factor experimental design,this paper aimed at investigating the effects of Aleppo Gall and the union NaF on lactate dehydrogenase of streptococcus mutans at different states.Methods Culture the streptococcus mutans at the states of planktonic and biofilm separately with brain-heart infusion(BHI) containing 2% glucose.Add Aleppo Gall,NaF and Aleppo Gall union NaF which were grouped according to factor experimental design to BHI and culture anaerobic for 18 hours.Assay the activity of 1octate dehydrogenase(LDH) with the pyruvate-dependent oxidation of NADH.The data were statistically analyzed by SPSS 17.0 software package.Make one way ANOVA of the inhibition rate of LDH.Results The activity of LDH at the state of biofilm was higher than that at the state of planktonic;Aleppo Gall,NaF and Aleppo Gall union NaF inhibited LDH both at the states of planktonic and biofilm;LDH in the state of planktonic was more sensitive with drug.The activity of LDH was statistically significant under different culture conditions;the inhibition rate of Aleppo Gall was the highest,the secend one was NaF and the last one was Aleppo Gall union NaF.Conclusions Aleppo Gall may inhibit acidogenic of streptococcus mutans by inhibiting the activity of LDH,then achieve the aim of anticariogenic.
