目的 研究陈醋对大鼠肾脏草酸钙结石形成的影响及可能的作用机制.方法 40只6~8周龄雄性SD大鼠随机分为5组,分别为对照组、模型组和3个陈醋(低、中、高剂量)干预组,每组8只.使用乙二醇和氯化铵诱导法建立大鼠肾脏草酸钙结石模型.28d后检测各组大鼠肾脏草酸钙晶体的形成情况及24 h尿液成石危险成分的含量,并通过免疫组织化学方法检测肾脏氧化应激损伤分子标志物的表达变化.结果 与模型组比较,中、高剂量陈醋干预组大鼠的肾脏晶体数量和24h尿草酸明显减少(P<0.05),且其肾组织内8-羟化脱氧鸟苷的表达明显降低(P<0.05),而超氧化物歧化酶的表达则明显升高(P<0.05).结论 陈醋可以明显抑制乙二醇和氯化铵诱导的大鼠肾脏草酸钙晶体的形成,其机制可能与陈醋影响大鼠草酸的代谢和降低大鼠肾组织氧化应激损伤有关.
Objective To investigate the effect and its mechanism of mature vinegar on the renal calcium oxalate crystal formation in rats.Methods Forty 6-8 weeks old SD rats were divided into 5 groups :control group, model group and vinegar intervention groups (low, middle and high dosage) randomly.The model of calcium oxalate stone was established by freely drinking water with 1% ethylene glycol and 1% ammonium chloride.Twenty-eight days after molding, the calcium oxalate crystal formation and risk factors of stone forming in 24h urine were detected.In addition, the expressions of oxidative stress injury related molecular markers were detected by immunohistochemistry.Results Compared to the model group, the crystal number and urinary oxalate were significantly reduced in middle and high dose vinegar intervention groups.The expression of 8-OHdG was significantly decreased, whereas the expression of SOD was significantly increased.Conclusions Mature vinegar could inhibit the renal calcium oxalate crystal formation induced by ethylene glycol and ammonium chloride in rat, which was related to the metabolism of urinary oxalate and reducing the renal oxidative stress injury.
