目的:通过观察脂肪酸合成关键酶FAS mRNA和脂肪酸分解关键酶ATGL mRNA表达的变化,探讨高住高练对肥胖大鼠脂肪酸代谢的影响。方法:适应性训练肥胖SD大鼠130只,分成13组:0周组,低住低练1、2、3、4周组,低氧安静1、2、3、4周组,高住高练1、2、3、4周组。低氧安静1、2、3、4周组大鼠每天在低氧下(氧浓度为13.6%,约相当于海拔3500 m高度)生活,自由活动,不训练,分别持续1周、2周、3周、4周;低住低练组在常氧下生活训练,高住高练组在低氧下生活训练。训练组大鼠采用水平动物跑台进行有氧运动,训练强度常氧下25~26 m/min,低氧下20~21 m/min,持续运动1 h/d,6 d/w,分别持续1、2、3、4周。相应周后,取肝脏和附睾脂肪,采用荧光定量PCR测试大鼠肝脏FAS mRNA和附睾脂肪ATGL mRNA表达。结果:(1)高住高练组大鼠肝脏FAS mRNA 1~4周与0周比较,各组均无显著性差异。与低住低练组比较,低氧安静和高住高练第3周显著低于低住低练(P〈0.05,P〈0.01)。(2)高住高练组大鼠脂肪组织ATGL mRNA第2、3周显著高于0周和低住低练相应周(P〈0.01),高住高练3周大鼠脂肪组织ATGL mRNA显著高于低氧安静(P〈0.05)。结论 :3500 m高住高练3周较低住低练降低肥胖大鼠肝脏FAS mRNA、增加脂肪组织ATGL mRNA表达,提示3500 m高住高练3周较低住低练抑制肥胖大鼠脂肪酸合成,促进脂肪酸分解。
objective To investigate the effects of living high and training high on fatty acid metabolism in obese rats. Methods One hundred thirty Sprague-Dawley obese rats were assigned into following groups:sedentary normoxic living(0 week),sedentary hypoxic living(1 to 4 weeks),normoxic training(1 to 4 weeks),and hypoxic training(1 to 4 weeks). Rats in the training groups exercised 1 hour per day,6 days per week for 4 weeks on a treadmill at a speed of 25-26 m/min under normoxic conditions or 20-21 m/min under hypoxic conditions,respectively. The expressions of liver FAS and adipose ATGL were tested by real-time quantitative PCR before and at the 1st,2nd,3rd,and 4th week of training. Results The liver FAS mRNA of hypoxic living and training group,and of sedentary hypoxic living group at the 3rd week were significantly lower than that of normoxic living and training group(P 〈0.05 and P 〈 0.01). The adipose ATGL mRNA of hypoxic living and training group at the 2nd and 3rd week was higher than that of baseline and that of normoxic living and training at the 2nd and 3rd week(P 〈 0.01,P 〈 0.05,P 〈 0.01). Conclusion Compared to exercise under normoxic conditions,exercise under hypoxic conditions for 3 weeks resulted in greater inhibition of fatty acid synthesis and greater promotion of lipolysis in obese rats.
