目的探讨基质金属蛋白酶-14（MMP-14）在骨关节炎（0A）患者滑膜、滑液中的表达及意义。方法采用半定量RT-PCR检测32例0A患者（实验组）及26例非0A患者（对照组）关节滑膜组织中MMP-14mRNA的表达水平；采用ELISA法检测关节滑液中MMP-14蛋白的表达水平。结果经RT-PCR检测MMP-14mRNA在实验组与对照组患者滑膜组织中的表达差异有统计学意义（P〈0．05），ELISA检测两组滑液中MMP-14蛋白表达差异有统计学意义（P〈0．05）。结论MMP-14在OA滑膜中呈高表达，表明在0A的发生及发展过程中可能起促进作用；在滑液中低表达，表明0A发生时MMP-14未大量释放至滑液，间接表明MMP-14的主要功能于滑膜细胞中表达并促进骨关节炎的发展。

Objective To evaluate the expression and clinical significance of matrix metalloproteinase 14（MMP-14） in osteoarthritis（OA） patients in synovium and synovial fluid of knee joint. Methods Semi-quantitative RT-PCR were used to detect the expression of MMP-14 mRNA in 32 case of OA （experimental group）and 26 non-OA patients（control group）synovium; protein level expression of MMP-14 in synovial fluid were analyzed by ELISA. Results The expression of MMP-14 mRNA in experimental group synovium of knee joint was significantly increased compared with control group（P〈0.05） ;but the protein level of MMP-14 was low in experimental group synovial fluid compared with control group（P〈0.05）. Conclusion The expression of MMP-14 is increased in OA patients synovium and maybe promote the progress of OA directly; the protein level of MMP-14 is decreased in OA synovial fluid,indicated the function of MMP-14 in OA synoviocyte indirectly.
