目的:研究脑通复方制剂对血管性痴呆大鼠学习记忆及胆碱酯酶活性的影响,探讨其对血管性痴呆治疗的作用及机制。方法:采用改良四血管法建立血管性痴呆模型;Morris水迷宫进行行为学测试,Ellman比色法测定海马匀浆中乙酰胆碱酯酶、丁酰胆碱酯酶活性。结果:脑通复方制剂能提高大鼠学习记忆成绩,降低海马乙酰胆碱酯酶活性。结论:脑通复方制剂对血管性痴呆大鼠学习记忆能力下降有明显的保护作用,其作用机制可能与抑制乙酰胆碱酯酶活性作用密切相关。
OBJECTIVE To investigate the effect of Nao Tong compound preparation on leraning, memory and activty of cholinesterase(ChE) in hippocampus tissue of vascular dementia (VaD) rats. METHODS The VaD model was created with the reformed Pulsinelli 4 - VO method. After establishing the models, the rats were treated with Nao Tong compound preparation. The abilities of learning and memory of the VaD rats were assessed by Morris Water Maze Behavior Test. The activities of ace- tylcholinesterase (ACHE) and butyrylcholinesterase (BuChE) in hippocampus tissue of vascular dementia rats were detected by the improved Ellman's colorimetric methods. RESULTS Compared with modle group, Nao Tong compound preparation could improve the learning and memory ability of rats,decrease the activity of ACHE. CONCLUSION Nao Tong compound prepara- tion has significant protective effect against the decrease of learning and memory of vascular dementia rats: the mechanism of its protectives effect may relate to the decrease activity of ACHE.
