不同分离纯化方法对草菇多糖抗氧化活性的影响

采用乙醇分步沉淀法、超滤法、柱层析分离纯化草菇多糖,并对其抗氧化活性进行研究。采用超滤法纯化草菇多糖可以增强抗·OH和DPPH·能力。超滤法获得分子量(MW〉100 ku)与(MW5 ku~10 ku)草菇多糖对·OH和DPPH·清除活性均高于未分级草菇多糖,对·OH清除活性分别为68.02%、48.41%,对DPPH·清除活性分别为95.50%、95.54%。采用乙醇分步沉淀法纯化草菇多糖可以增强清除·OH能力。乙醇分步沉淀获得VWPF60、VWPF80草菇多糖对·OH清除活性较高,分别为72.26%、71.77%。

英文摘要:

The ethanol fractional precipitation, ultrafiltration, column chromatography were used to separate and purify the polysaccharides from V. volvacea, and their antioxidant activity was studied. The activity against hydroxyl radicals and DPPH was improved when V. volvacea polysaccharide was purified by ultrafiltration. Using ultrafiltration, two kinds of polysaccharides with molecular weight (MW 100 ku) and (MW 5 ku-10 ku) were obtained, which had stronger scavenging activity against hydroxyl radicals and DPPH than the unrated polysaccharides. The scavenging activity against hydroxyl radicals respectively were 68.02%, 48.41%, and which against DPPH were 95.54% and 95.50%. By using ethanol precipitation step purification V. volvacea polysaccharide, the scavenging activity against hydroxyl radicals was improved. The VWPF60, VWPF80 of V. volvacea polysaccharide by ethanol fractional precipitation had stronger scavenging activity against hydroxyl radicals, which were 72.26%, 71.77%, respectively.
