在大肠杆菌中共表达DJ-1(PARK7)蛋白、小肽和-αSyn,研究DJ-1影响小肽抑制α-Syn(Α53T,S129Α)聚集的效果.以文献报道的效果最好的小肽为对照,构建4种小肽(Pn:P1-P4)与DJ-1共同表达的双顺反子,共表达融合蛋白α-Syn-GFP;利用融合蛋白的折叠在细菌中可视化技术研究DJ-1协同小肽Pn抑制α-Syn(Α53T、S129Α)聚集的效果.发现:DJ-1与小肽P1(MRGVVΑΑΑR)的共同作用,能显著抑制α-Syn(S129A)的聚集,其效果提高了29%;对于α-Syn(A53T),在DJ-1的协同作用下,小肽P3(MRVGGΑVVTGR)抑制α-Syn(A53T)聚集的效果提高了134%,小肽P4(MRVGGΑVVR)提高了129%.结论表明,在大肠杆菌中,DJ-1与小肽的相互作用可以不同程度的抑制α-Syn(Α53T,S129Α)的异常聚集.
