目的研究艾灸对衰老大鼠组织细胞周期及PKC、PP2A表达的影响。方法以注射D一半乳糖溶液法制备亚急性衰老大鼠模型,采用艾灸“肾俞”穴进行治疗,并与模型组和正常组进行对照,以流式细胞术和免疫组化法观察各组大鼠肝脏组织细胞周期及PKC、PP2A表达的变化。结果与正常组相比,亚急性衰老模型大鼠组织PKC、PP2A阳性表达面积和阳性表达光密度均明显降低(均P〈0.01);艾灸组PKC阳性表达面积和阳性表达光密度均较亚急性模型组显著增高(P〈0.05,P〈0.01),PP2A阳性表达面积与阳性表达光密度也有显著增高(P〈0.01,P〈0.05)。亚急性衰老模型大鼠组织G0/G1期细胞比例较正常组升高(P〈0.05),PI指数降低(P〈0.05);而艾灸组G0/G1期细胞比例则较模型组有所下降,P1指数有所升高,但两组间未见统计学差异(P〉0.05)。结论艾灸可能是通过调节衰老机体组织PKC、PP2A活性,降低G0/G1期细胞比例,升高P1指数,从而达到延缓衰老的目的。
Objective To investigate the effect of moxibustion on cell cycle and PKC and PP2A expressions in tissues of senescent rats. Methods Subacute aging rats model were established by injection with D-gal solution. Points Shenshu ( BL 23 ) were treated with moxibustion in treatment group, contrasting with a model group and a normal group. Flow cytometry (FCT) was used for the change of cell cycle and immunohistochemistry (IHC) was used for the expressions of PKC and PP2A in liver tissues. Results In comparison with normal rats, the positive-expressed areas and photodensity of PKC and PP2A in subacute aging model group were all significantly lower( P 〈 0. 01 ). In the moxibustion group, the positive-expressed areas and photodensity of PKC and PP2A were signifi- cantly higher than that in subacute aging model group respectively ( P 〈 0.05, P 〈 0.01/P 〈 0.01,P 〈 0.05 ). The cell percentage at GO/G1 phase in liver tissues of subacute aging rats was decreasd and the index number of PI was increased significantly( P 〈 0.05 ), whereas, in the moxibustion group, the cell percentage at GO/G1 phase was higher than that of model group and the index number of PI was lower, but there was no difference between the model group and moxibustion group(P 〉 0.05 ). Conclusion These results suggest that Moxibustion probably regulates PKC and PP2A activity of senile rats, reduces the cell percentage at GO/G1 phase, increases the index number of PI, and hence to delay senescence.
