目的:以基因芯片技术从差异基因表达角度分析骨关节炎肾阳虚证与信号转导相关差异表达基因.方法:先后筛选出符合西医膝骨关节炎和中医肾阳虚证诊断标准的患者两组,每组8例,与8例正常对照组比较.两组病人均取血、提取mRNA,经标记、杂交、清洗、芯片扫描、图像的采集与数据分析等过程,分别混合与对照组杂交作两张基因芯片,最后对数据进行信息挖掘.结果:两张芯片各获得162和75条差异表达基因,其中与信号转导相关的分别为26条和11条;两张芯片共同差异表达基因6条,与信号相关的有3条,占总数的1/2.提示:信号转导是生命活动的一种最基本和最重要的方式.细胞间信号转导异常可能在肾阳虚证骨关节炎的发生、发展过程中起着重要作用.
Objective: To analyze the differentially expressed genes associated with signal transduction of patients who suffered from Osteoarthritis of the deficiency of kidney - Yang through the technology of cDNA microarrays. Methods: We select two groups of patients who fulfilled the diagnosis and included criteria of osteoarthritis of the deficiency of kidney - Yang, each group includes 8 patients, at the same time, 8 health peoples are selected as control group. We obtain differentially expressed genes by drawing blood, extracting mRNA, labeling, hybridizing, scanning, data analyzing and so on. At last, we select differentially expressed genes associated with signal transduction to analyze it. Results: 162 and 75 differentially expressed signal transduction genes are acquired in two cDNA microarrays, in them, 26 and 11 genes associated with signal transduction are obtained respectively. The same 6 genes differentially expressed both in two cDNA microarrays have been selected. Discussion: Signal transduetion is a basic and important way of life activity, and it plays an important role in occurrence and development of osteoarthritis of the deficiency of kidney - Yang.
