探讨并比较了苦参、半枝莲、白花蛇舌草的单一提剂水/醇提物及其组合剂对人结肠癌细胞株SW620生长的抑制作用.通过体外培养SW620细胞,加入终浓度分别为1、3、5mg.mL-1的单一提剂及其组合剂,采用MTT法测定药物对SW620的增殖抑制率(IR).结果表明单一提剂对SW620体外增殖均有抑制作用,且呈浓度剂量依赖性;中草药醇提剂抑瘤作用均强于水提剂.组合剂中,除半枝莲与白花蛇舌草水提组合剂比单一提剂作用弱外,其余组合剂均强于单一提剂的抑瘤率,且呈浓度剂量依赖性;中草药醇提液组合剂抑瘤作用均强于水提液.说明苦参、半枝莲、白花蛇舌草水/醇提取物对SW620细胞的生长均有抑制作用,醇提剂抗肿瘤有效成分多于水提剂;三种中草药组合剂抗肿瘤效果优于单一提剂(除半枝莲与白花蛇舌草水提组合剂外).
Flavescens,Scutellaria barbata and Hedyotis diffusa are three commonly used anti-cancer Chinese medicinal herbs.The present study investigated the inhibitory effect of water or alcohol extracts as well as combination of extracts of three herbs in vitro,using human colon cancer cell line SW620 as an experimental model.The MTT assay was used to quantify the proliferation of the cancer cell.The water and alcohol extracts of all three herbs all had inhibitory effect on the cancer cells in a dose-dependent manner.The alcohol extracts all had a higher inhibitory effect than water extracts.Various combinations of two water extracts had higher inhibitory effect than single water extract alone,except the combination of the water extracts from Scutellaria barbata and Oldenlandia diffusa.All various combinations of two alcohol extracts all had higher inhibitory activity than their water extract counterparts.In conclusion,all extracts of three herbs can significantly inhibit cancer cells proliferation.Ethanol extracts are more potent than aqueous extracts.In most cases,combination of two extract show synergistic anti-cancer effect.
