目的探讨地皮菜水应激蛋白(water stress proteins,WSP1)对结肠癌DLD1细胞-细胞、细胞-基质之间黏附的影响和分子机制。方法结肠癌细胞DLD1经WSP1处理后,利用细胞黏附实验检测DLD1细胞-细胞、细胞-基质之间黏附的影响。利用real time PCR和Western blot技术检测DLD1细胞内E-cadherin、N-cadherin、vimentin和integrinβ1 mRNA和蛋白的表达情况。结果经WSP1处理DLD1细胞后,DLD1细胞-细胞黏附率升高(P〈0.01),细胞-基质黏附率降低(P〈0.01),E-cadherin表达水平升高,N-cadherin,vimentin和integrinβ1表达水平降低。结论 WSP1能够促进DLD1细胞间黏附并降低细胞-基质间黏附,进而抑制结肠癌细胞的侵袭和浸润。
Objective To explore the effect of water stress proteins (WSP1) from Nostoc commune Vauch. on the adhesion of colon cancer DLD1 cell and its molecular mechanisms. Methods DLD1 cells were treated with WSP1. Cell adhesion assay was applied to detect the effect of WSP1 on the cell-cell and cell-matrix adhesion of DLD1 cells. The mRNA and protein expression levels of E-cadherin, N-cad- herin, vimentin and integrin β1 were detected by real time PCR and Western blot. Results WSP1 elevated the DLD1 cell-cell adhesion rate(P 〈0. 01 ), decreased cell-matrix adhesion rate notably(P 〈 0. 01 ). After WSP1 treatment, the E-cadherin expression increased, and the N-cadherin and vimentin expression reduced. Furthermore, the expression of integrin β1 was attenuated by WSP1 treatment. Conclusion WSP1 could inhibit the invasion of DLD1 cells through promoting cell-cell adhesion and reducing cell-matrix adhesion.
