目的观察2型糖尿病时大鼠牙龈组织中DPP—Ⅳ分布和表达情况。方法将实验动物分为高脂糖饮食组、2型糖尿病组和对照组,以高脂糖饮食加链脲霉素腹膜内注射制备2型糖尿病动物模型,根据葡萄糖耐量试验异常确定2型糖尿病动物模型成功。采用免疫组化染色法观察二肽基肽酶Ⅳ(DPP-Ⅳ/CD26)在肠、肾和牙龈中的分布和表达。结果DPP-Ⅳ主要定位于肠上皮、肾小管细胞和牙龈黏膜上皮细胞。在肠和肾组织无论高脂糖饮食组(HF)还是2型糖尿病模型组(DM)DPP-Ⅳ表达均强于对照组。在牙龈组织,DM组大鼠强阳性表达,HF组大鼠弱阳性表达,对照组大鼠阴性表达。结论在2型糖尿病时,DPP—Ⅳ/CD26在大鼠牙龈组织中出现高表达。
Objective To observe expression of DPP-Ⅳ in intestine, kidney and gingival of the rats with Type Ⅱ diabetes. Methods Three groups of rats ( control ,high fat diet (HF) and HF plus STZ for Type Ⅱ diabetes mellitus (DM) ) were monitored with plasma glucose and insulin for 12 weeks. Type Ⅱ diabetic rats were confirmed by the glucose/insulin tolerance test. The expressions of DPP-Ⅳ in intestine, kidney and gingival were examined using immunohistochemistry. Results Compared with rats on the normal diet, the expression of DPP-Ⅳ was significantly increased in the intestine, kidney and gingival in rats with Type Ⅱ diabetes. The rats with high fat diet also moderately upregulated DPP-Ⅳ expression in intestine ,kidney and gingival. Conclusion These findings suggested that the overexpression of DPP-Ⅳ on gingival in Type Ⅱ diabetes could affect health of periodontal tissues .
