目的:检测人参复合物制剂DS1226对人乳腺癌细胞细胞ZR-75-30细胞增殖、凋亡和运动的影响。方法:体外培养人乳腺癌细胞ZR-75-30,予DS1226干预组为DS1226组(实验组),无药物组为对照组。CCK8检测DS1226对ZR-75-30细胞增殖的影响,流式细胞术检测细胞凋亡和细胞周期的改变,进一步用细胞划痕实验检测划痕愈合率。结果:与对照组相比较,CCK8结果显示DS1226时间依赖性抑制人乳腺癌细胞ZR-75-30的细胞增殖,24 h、48 h和72 h的细胞增殖抑制率分别是3.01±0.12%,13.10±0.51%和23.66±4.85%(P〈0.01);FACS结果显示DS1226干预组的凋亡率从干扰前3.20±0.14%增强至药物刺激后的7.17±0.32%(P〈0.05);划痕实验显示DS1226干预组24 h与对照组相比划痕愈合明显下降(P〈0.01)。结论:人参复合物制剂DS1226能显著抑制人乳腺癌细胞ZR-75-30的细胞增殖、增强细胞凋亡和降低细胞运动能力。
Objective: To study the effect of Ginseng compound DS1226 on proliferation, apoptosis, and migration of human breast cancer cell line ZR-75-30. Methods: Cultured ZR-75-30 cells were treated with DS1226 as trial group and without DS1226 as control group. Cell proliferation was determined by CCK8 assay,cell apoptosis and cell cycle were analyzed by FACS, and cell migration was evaluated by scratch assay. Results:Compared to control group, DS1226 significantly decreased the proliferation of ZR-75-30 cells in a time-dependent manner(P〈0.01), induced cell apoptosis(P〈0.05), and reduced the rate of wound closure(P〈0.01).Conclusion: DS1226 inhibits cell proliferation and migration, and enhances cell apoptosis in human breast cancer cell line, ZR-75-30.
