在获得鱼类淋巴囊肿病毒(lymphocystis disease virus,LCDV)单克隆抗体的基础上,建立了淋巴囊肿病毒的免疫电镜(IEM)和斑点免疫印迹(Dot—blot)诊断方法。应用免疫电镜方法检测样品,观察结果显示:胶体金颗粒集中结合在淋巴囊肿病毒粒子囊膜周围,在病毒外区域没有胶体金颗粒散布,整个视野背景清洁,无散在的金颗粒和其他污染物;斑点免疫印迹诊断结果显示:在使用EDTA抑制内源酶后,所检测样品发色后呈明显褐色,呈现明显的LCDV阳性反应,而设立的两组阴性对照结果均不发色;诊断结果说明所建立的淋巴囊肿病毒单克隆抗体诊断技术灵敏、稳定、特异、可靠。
In the present study, the Monoclonal antibodies against lymphocystis disease virus (LCDV) were used to development immuno - eleetrton microscopy test (IEM) and immuno - dot blot assay ( Dot - blot). Results of immuno - electrton microscopy test showed that the high density gold particles were located at the outermost surface of freshly purified virus particles, but not the viral nueleocapsid or outside the virions. Very little background labeling was observed. In immuno - dot blot assay EDTA was the most effective of all the reagents tested for inhibition of endogenous enzyme contained in samples. The reaction with treated samples was the strongest brown color. The results of the experiments indicated that these two kinds serological techniques were high sensitivity, stability, validated when were used to diagnose lymphocystis disease virus of fish that collected from marine culture farm.
