舒腰合剂抗大鼠椎间盘软骨终板老化的作用

目的评估舒腰合剂抗老龄雌性大鼠椎体终板老化的作用。方法采用雌性22月龄老年大鼠50只,随机分为舒腰合剂大、中、小剂量组、雌激素组及老年对照组,另6月龄大鼠12只作为青年对照组,连续给药45d。常规组织切片,HE染色,测量椎体软骨终板血管芽相对面积及非钙化层/钙化层比值,组间比较采用单因素方差分析。结果软骨终板血管芽相对面积测量显示,与老年对照组(24.07±8.80)相比,舒腰合剂大剂量组增加约54%,青年对照组增加约74%,而中、小剂量组、雌激素组虽各有不同程度的增加,但均无统计学意义(P〉0.05)。软骨终板非钙化层/钙化层厚度比值测量显示,与老年对照组(0.82±0.23)相比,舒腰合剂大剂量组增加约37%,青年对照组增加约48%,具有统计学意义(P〈0.05);而舒腰合剂中、小剂量组、雌激素组虽各有不同程度的增加,但均无统计学意义(P〉0.05)。结论大剂量舒腰合剂可延缓增龄过程中椎间盘软骨终板血管芽相对面积的减少及软骨终板非钙化层/钙化层比值的降低,提示可作为早期椎间盘退变的预防用药。

英文摘要:

Objective To evaluate the therapeutic effect of Shuyao Mixture on the aging of vertebral endplate in rats. Methods Sixty-two female rats were divided into two groups: the 22-month group (50) and the 6-month young group (12). The 50 rats were randomly further divided into high-dosage Shuyao Mixture group (4.5 g/kg), medium-dosage group (1.8 g/kg), low-dosage Shuyao Mixture group (0.72 g/kg), aged negative control group, and estrogen group. The 12 young rats received Shuyao Mixture for consecutive 45 days. Histological sections were used to evaluate the relative area of vascular buds in the endplate, and the ratio of un-calcified layer to calcified layer in the endplate was analyzed by Square's methods. One-way ANVOA was used to analyzed the values. Results Compared with that in the aged control group, the relative area of vascular buds increased by 54% in high-dose group, 74% in young group, but there was no significant increase in the medium- and low-dosage groups and estrogen group (P〉0.05). The ratio of un-calcified layer to calcified layer increased by 37% in high-dose group, 48% in young group (P〈0.05), but there was no significant increase in the medium- and low-dosage groups and estrogen group (P〉0.05). Conclusion Shuyao Mixture can enhance the relative area of vascular buds and the ratio of un-calcified layer to calcified layer in endplate, which suggests that Shuyao Mixture may prevent early degeneration of intervertebral disc.
