目的研究养心通脉方对异丙肾上腺素诱导的大鼠心肌缺血损伤模型心肌细胞线粒体呼吸功能的影响。方法实验大鼠被随机分为3组(每组15只):对照组、模型组、养心通脉方组,实验结束后所有动物采用脱颈椎法处死,通过测定呼吸控制率(RCR)、二磷酸腺苷磷/氧(ADP/O)比值及ATP的浓度来评价线粒体的呼吸功能,并对线粒体内胆固醇、磷脂(PL)、甘油三酯(TG)和游离脂肪酸(FFAs)的含量进行测定。结果模型组中大鼠心肌细胞线粒体内RCR、ADP/O比值及ATP浓度均明显低于对照组(P<0.001),而在养心通脉方组中心肌细胞线粒体内RCR、ADP/O比值及ATP浓度均接近于对照组(P>0.05)。与对照组相比,模型组中胆固醇、TG和FFAs的水平均明显升高(P<0.001),同时磷脂(PL)水平明显降低(P<0.001)。但在养心通脉方组中心肌细胞线粒体内胆固醇、TG、FFAs以及PL的水平均与对照组无明显差异(P>0.05)。结论养心通脉方对异丙肾上腺素诱导的大鼠心肌缺血损伤模型心肌细胞线粒体的呼吸功能及其膜完整性具有保护作用。
Objective To evaluate the effect of Yangxin Tongmai Fang on mitochondrial respiration of cardiomyocyte in the isoproterenol-induced myocardial ischemia injury rats model. Methods The experimental rats were divided into three groups:control group, model group, Yangxin Tongmai Fang group, 15 rats in each group. After the experimental period, the animals were sacrificed by cervical decapitation. The mitochondrial respiration of cardiomyocyte was evaluated by the levels of respiratory control ratio (RCR), ADP/O ratio and ATP concentration. The mitochondrial cholesterol, phospholipids (PL), triacylglycerol(TG) and free fatty acids (FFAs) were also determined. Results The levels of RCR, ADP/O ratio and ATP concentration in model group were lower than that in control group (P<0.001), the RCR, ADP/O ratio and ATP concentration in Yangxin Tongmai group were close to the control group (P>0.05). Compared with the control group, the levels of cholesterol, TG and FFAs in model group were significantly increased (P<0.001), and the PL content was decreased obviously (P<0.001). But there were no significant difference in the levels of cholesterol, TG, FFAs and PL between Yangxin Tongmai Fang group and control Group (P>0.05). Conclusion Yangxin Tongmai Fang shows protective effect on mitochondrial respiration and its membrane integrity of cardiomyocyte in the isoproterenol-induced myocardial injury rat models.
