目的探讨HtrA1(high temperature requirement A1,HtrA1)和TGF-β1(transforming growth factor-beta 1,TGF-β1)在妊娠期高血压病患者(简称妊高病)胎盘组织中的表达及作用。方法利用免疫组化SABC法检测正常妊娠者60例(对照组)和妊高病者70例(实验组)(其中妊娠期高血压组24例,轻度子痫前期组20例,重度子痫前期组26例)胎盘组织中HtrA1和TGF-β1的蛋白表达,并进行相关性分析。结果 HtrA1在实验组中的表达较对照组显著增高(平均灰度分别为155.05±3.13、152.69±4.11,P〈0.01);与对照组比较,轻度子痫前期组、重度子痫前期组均显著增高(分别为154.05±4.96、156.03±5.07,P分别〈0.05、〈0.01)。TGF-β1在实验组中的表达较对照组显著增高(平均灰度分别为156.33±4.79、152.39±4.84,P〈0.01);与对照组比较,轻度子痫前期组、重度子痫前期组均显著增高(分别为159.79±3.92、165.82±3.20,P〈0.01)。Pearson相关分析HtrA1与TGF-β1的表达水平在实验组中呈正相关。结论 HtrA1和TGF-β1可能通过某些机制相互调控、协同,两者高表达可能与妊高病的发生发展有关
Objective To study the expressions of high temperature requirement A1(HtrA1) and transforming growth factor-beta 1(TGF-β1) in placenta of patients with hypertensive disorder-complicating pregnancy and their relationship.Methods We recruited 60 normal pregnant women(control group) and 70 women with hypertensive disorder-complicating pregnancy(study group)(including 24 cases of gestational hypertension,20 cases of mild preeclampsia and 26 cases of severe preeclampsia).The expressions of HtrA1 and TGF-β1 in placenta in each group were detected by immunohistochemical SABC methods.The relationship between HtrA1 and TGF-β1 was determined.Results The level of HtrA1 expression was significantly higher in the study group than in the control group(the mean grey number was 155.05±3.13 and 152.69±4.11,respectively;P〈0.01).Compared with that in the control group,the mean grey numbers of mild preeclampsia and severe preeclampsia were both significantly higher(154.05±4.96 and 156.03±5.07,respectively;P〈0.05,P〈0.01).The level of TGF-β1 expression was significantly higher in the study group than in the control group(the mean grey number was 156.33±4.79 and 152.39±4.84,respectively;P〈0.01).Compared with that in the control group,the mean grey numbers of mild preeclampsia and severe preeclampsia were both obviously higher(159.79±3.92 and 165.82±3.20,respectively;both P〈0.01).Pearson correlation analysis showed a positive correlation between HtrA1 and TGF-β1 expressions in the study group.Conclusion HtrA1 and TGF-β1 may regulate and synergize with each other.The high expressions of HtrA1 and TGF-β1 may be related to the pathogenesis and development of hypertensive disorder-complicating pregnancy
